Nomenclature: C57BL/6-Piezo2tm1.1Tac

  • The model carries a conditional knock-in point mutation (cKI-PM) in the Piezo 2 (piezo-type mechanosensitive ion channel component 2).
  • In absence of Cre, the model is expected to express the wildtype Piezo 2 (Fam38b) allele.
  • Upon crossbreeding with a Cre model, the constitutive KI-PM allele may be obtained, facilitated by deletion of the wildtype exon 54 and expression of the mutated Piezo 2 ED2799D protein.
  • This model might be useful for the study of Distal arthrogryposis (DA) disease progression and understanding.

Origin

The Piezo 2 mouse was developed at Taconic Biosciences on behalf of Scripps Health in 2012. The model was generated using BAC clones from the C57BL/6J RPCIB-731 BAC library and was transfected into the Taconic C57BL/6NTac ES cell line. Homologous recombinant clones were isolated using double positive (NeoR and PuroR) selection. The conditional KI-PM allele was obtained after Flp-mediated removal of the selection markers. Taconic received donor animals from Scripps in 2023 and the model is stored as homozygous sperm.
