Overview

Nomenclature: B6;C3-Tg(CAG-DPP4)52Ctkt

  • Mice expressing human DPP4 are susceptible to infection by MERS-CoV, the virus that causes Middle East respiratory syndrome coronavirus. The hDPP4 mouse model expresses human DPP4 cDNA under the control of the CAG promoter.
  • Published reports indicate that the hDPP4 mouse develops disseminated, fatal disease upon infection with MERS-CoV at higher viral doses
  • This model is useful for study of MERS disease pathogenesis, treatment and vaccines
  • This model may also be useful to test pan-coronavirus vaccines
  • The genetic background of this model will change over time as it will be continually backcrossed to C57BL/6NTac. Wild type (wt/wt) mice from the 18517 colony are the recommended wild type control at this time.
  • Distribution of this model is subsidized by CEPI

Orders by weight: Taconic cannot accept orders by weight for this model. Please note that shipments may contain animals with a larger weight variation.

Origin

The hDPP4 Mouse was generated in the laboratory of Chien-Te (Kent) Tseng at University of Texas Medical Branch (UTMB) in Galveston.  The transgene, comprising CAGG enhancer/promoter, intron sequence, human DPP4 cDNA, and rabbit beta globin poly(A) signal was injected into B6C3F1/J x C57BL/6J zygotes. Taconic maintains the line by mating hemizygous hDPP4 mice to C57BL/6NTac wild type mice.

Genetics

Species: Mouse

Other Genetic Notes:

The genetic background of this model will change over time as it will be continually backcrossed to C57BL/6NTac. Wild type (wt/wt) mice from the 18517 colony are the recommended wild type control at this time.

Data

The hDPP4 Mouse can be used to assess MERS therapeutics

hDPP4 data

Fig 1. hDPP4 mice were infected with 104 TCID50 of MERS-CoV and then treated 12 h later with either therapeutic antibody m336 or a control antibody m102.4. Mice were monitored daily for A) bodyweight change and B) morbidity and mortality.
