Nomenclature: STOCKTg(Prnp-MAPT*P301L)JNPL3Hlmc

  • Carries the transgene for the human P301L mutation of the microtubule-associated protein tau gene (MAPT) and expresses mutant human TAU protein
  • Aggregates of filaments of TAU result in Neurofibrillary tangles (NFT) which are associated with Alzheimer's disease, Pick disease and other neurological syndromes
  • Develops behavioral and motor disturbances related to development of NFT
  • Model 2508 is homozygous for the transgene and develops a phenotype that mimics human tauopathies in a more rapid time frame than hemizygous mice
  • Useful for studying Alzheimer's disease, Pick's disease and other neurological syndromes associated with NFT
  • This model is on a C57BL/6, DBA/2, SW mixed background.
  • The appropriate wild type control stock is model #18392.
  • Pink eyed animals, associated with certain coat colors, and the Pde6brd1 retinal degeneration mutation can cause light sensitivity and/or blindness in some animals. This may impact the results of behavioral testing. Upon request, animals can be screened for eye color, coat color, and/or rd1 homozygosity for an additional fee.

Origin

The Tau mouse was developed in the laboratory of Mike Hutton at the Mayo Clinic. The model was created by microinjecting transgenic constructs containing the P301L mutation of the microtubule associated tau gene and mouse prion promoter into B6D2F1 x SW zygotes. The resultant mice from Founder Line JNPL3 were mated to B6D2F1. Taconic received stock in December 2000. The mice were derived by embryo transfer and mated to SW and B6D2F1 hybrids and embryo transfer derived. Offspring from both backgrounds were combined and were mated to B6D2F1 mice to produce hemizygous and wild type mice. Hemizygous Tau mice were intercrossed to generate homozygous Tau mice. A genetic refresh was initiated in part to generate a suitable wild type control stock. Homozygous 2508 Tau females were mated to Swiss Webster males. Hemizygous males from this cross were mated to B6D2F1 females. Wild type offspring from this second cross were used to establish control model #18392, whereas hemizygous males and females were intracrossed to generate homozygous mice to re-establish model #2508. The colony is maintained by mating homozygous females and males.
