Model #21546 contains the allele shown as “targeted allele” above. Cre-mediated recombination generates the allele shown as “mutant allele” above. Deletion of exon 7 results in a frame shift and premature termination of translation at a stop codon in exon 8. Use of viral-mediated cre rather than cre driver mice is recommended to prevent in utero deletion of Prkn and developmental compensation.

Origin
The floxed Prkn allele was generated by Lexicon Pharmaceuticals. The targeting vector included Prkn exon 7 flanked by loxP-sites and an FRT-flanked neomycin selection cassette. Targeting ES cells were injected into blastocysts. Chimeric mice were bred to protamine-cre mice to generate heterozygous pups containing one floxed and one deleted allele. Taconic received stock from the laboratory of Ted Dawson at Johns Hopkins University.
